Manager Roberto Di Matteo revealed the news on Tuesday, adding that Ryan Bertrand will replace the 31-year-old at left-back.
"Ashley Cole won't be available tomorrow, he has a problem with his hamstring, shouldn't be out for long," he said.
"Ryan Bertrand has always performed well for us when he's played and I have full confidence he will play a great game."
Chelsea are currently second in Group E with four points from three matches, three points behind leaders Shakhtar.
