The 24-year-old managed to seal a 4-6 6-1 7-6(3) triumph, ensuring that two British women will be in the last 16 of the tournament for the first time following Heather Watson's progression.
Vesnina took the first set thanks to a break point in the seventh game, but Konta bounced back in the second to go an early break up and eventually draw level.
The Brit took a 4-3 lead in the third set, but Vesnina stayed in contention and the match was decided with a tie-break, which Konta took 7-3.
The Australian Open semi-finalist will face Romania's Monica Niculescu in the next round.
