The Venezuelan has not won a PGA event since 2011 but he has given himself a clear opportunity to get back on the board with an excellent showing on Friday.
The world number 244 hit four birdies on the front nine but coming in, he recorded an eagle on the 13th before hitting a hole-in-one on the par-three 17th.
American Hudson Swafford is in second position, with compatriot Michael Johnson a shot further behind in third place.
Six players - including former Major winner David Toms - sit in a share of fourth position, but Scotland's Martin Laird remains down the standings in 42nd spot.
