The West Indian all-rounder's contribution, which was achieved off just 66 deliveries, is a record in the Twenty20 format of the sport.
Tillakaratne Dilshan (33) and AB de Villiers (31 from eight balls) added to Gayle's score, which in turn saw the Challengers set a T20 record of 263-5 from their permitted overs.
The Warriors' reply stuttered from the start and they lost their opening four batsmen, including Englishman Luke Wright, for a combined total of 41 runs.
Aussie Steven Smith ended as their top scorer with 41 runs to his name, but it was Gayle who had the final say when he claimed two wickets in the final over as the Warriors fell well short of their victory target.
