He made short work of the rest of the field in his semi-final, quickly establishing a lead and coasting over the line in a time of 9.86s.
Bahrain's Andrew Fisher was disqualified from the race after a false start, while Andre De Grasse of Canada joined Bolt in automatically qualifying from the second semi-final.
Bolt's main rival for the gold medal Justin Gatlin also won his semi-final with ease, although he came through in a slower time of 9.94s.
Yohan Blake finished second behind Gatlin, while Jimmy Vicaut and Ben Youssef Meite were the automatic qualifiers from the first semi.
Great Britain's James Dasaolu and Chijindu Ujah both missed out on a place in the final, meanwhile.
